First-in-Human Trial of YCT-529 Shows Promise for Non-Hormonal Male Contraception

YourChoice Therapeutics has published results from a Phase 1a clinical trial that demonstrated YCT-529, a novel non-hormonal oral male contraceptive, to be safe and well tolerated in healthy men. Developed as a retinoic acid receptor-α antagonist, YCT-529 works by disrupting sperm production without affecting hormone levels or mood.

In the trial, 16 male participants received escalating single doses (10–180 mg) of YCT-529, with no adverse effects on heart rate, hormone levels, inflammatory markers, sexual desire, or mood. The study also found no significant impact from food intake on the drug’s absorption.

These results mark a major step forward in male contraceptive development. A follow-up Phase 1b/2a trial is now underway to evaluate the effects of repeated dosing over 28 and 90 days on sperm parameters.
